ELISA is one of the most commonly used assays in modern labs. However, if you need to measure multiple proteins the workload can quickly get very large. This is where ELISAGenie’s new Genieplex assay comes in. Genieplex is a multiplex ELISA technology which allows the researcher to measure up to 24 analytes from as little as 15µl of sample. These assays use beads, which have different sizes and fluorescence intensities, tagged to antibodies. These bead populations can then be distinguished into separate populations on a flow cytometer to separate the analytes.
Genieplex principle and protocol
When you start your Genieplex assay you will have different bead populations, each population with an antibody against a different protein in each well. You add as little as 15µl of sample, incubate for 1 hour, then add a biotinylated antibody cocktail against the proteins.
After a further 30 minute incubation Streptavidin-PE is added before the beads are run on a flow cytometer.
The different bead populations can be easily separated based on size and fluorescent intensity. Using the free software the concentration of protein in each sample can be calculated by comparing the PE fluorescent intensity of samples to that of the standards provided in the kit.
