HIGH PERFORMANCE GEL ELECTROPHORESIS FOR SERUM ANALYSIS WITH B1-B2 SEPARATION
TEST PRESENTATION
Protein electrophoresis is a well-established technique routinely used in clinical laboratories for screening samples for protein abnormalities (gammopathies, inflammatory pattern…). On the HYDRASYS instruments, utilizing the HYDRAGEL B1-B2 assay gels, serum proteins are separated into 6 major fractions:
- Albumin
- Alpha-1
- Alpha-2
- Beta-1
- Beta-2
- Gamma
The semi-automated HYDRASYS instrument offers sample application, electrophoretic migration and staining. The visualization of the gel provides qualitative analysis, while the reading of the agarose gels on a Sebia reader provides protein profiles for relative quantitative analysis. On the HYDRASYS 2 SCAN, the scanning system is embedded in the instrument.
Serum and urine protein electrophoresis are essential to detect and quantify monoclonal components for the diagnosis and monitoring of patients with monoclonal gammopathies (Multiple Myeloma, MGUS, Waldenström disease…).
The B1-B2 assay gels demonstrate better separation and identification of some monoclonal bands migrating in this zone.
TEST PRINCIPLE
The HYDRAGEL B1-B2 assay is based on the principle of electrophoresis separation on agarose gel.
The HYDRASYS system is a semi-automated multi-parameter instrument. The automated steps include processing of the agarose gel in the following sequence:
- Sample application
- Electrophoretic migration
- Drying
- Staining with amidoblack dye
- Destaining
- Final drying
The sample loading on the applicators can be automated using the ASSIST pipetting station.
PERFORMANCES
- Resolution: optimal focalization of the fractions obtained by applying a very thin sample’s application using the Sebia proprietary applicators
- Sensitivity:
- Internal sensitivity study demonstrated that a monoclonal protein can be detected in a serum at a concentration as low as 21 mg/dL. According to the mobility of the monoclonal component and polyclonal background, the sensitivity may vary.
- Full traceability and positive ID is obtained, by connecting the HYDRASYS to the ASSIST pipetting station
