HIGH RESOLUTION GEL ELECTROPHORESIS FOR SERUM, URINE OR CEREBROSPINAL FLUID (CSF) PROTEINS
TEST PRESENTATION
The Hydragel HR Protein electrophoresis is a well-established technique routinely used in clinical laboratories for screening samples for protein abnormalities (gammopathies, inflammatory pattern…).
Although most clinical laboratories are satisfied with the “traditional” five-zone pattern of serum proteins, high resolution techniques can yield additional diagnostic information. On HYDRAGEL 7 HR and HYDRAGEL 15 HR gels, serum, urine or cerebrospinal fluid (CSF) proteins separate up to ten fractions. Each fraction contains one or more proteins. The composition of the gel, the electrophoretic conditions and the choice of the stain allow an excellent resolution and a high sensitivity particularly in the gamma-zone.
The semi-automated HYDRASYS instrument offers sample application, electrophoretic migration and staining.
TEST PRINCIPLE
The HYDRAGEL 7 HR and HYDRAGEL 15 HR assay is based on the principle of electrophoresis separation on agarose gel.
The HYDRASYS system is a semi-automated multi-parameter instrument.
The automated steps include processing of the agarose gel in the following sequence:
- Sample application
- Electrophoretic migration
- Drying
- Staining
- Destaining
- Final drying.
The electrophoretic protein separations can be stained either with acid violet or amidoblack. The stained gels are evaluated visually for pattern abnormalities. The visual observations can be followed by densitometry to obtain semi-quantitative, relative values of the individual or combined protein fractions.
The sample loading on the applicators can be automated using the ASSIST pipetting station.

- Internal sensitivity study demonstrated that a monoclonal protein can be detected in a serum:
- On HR1 or HR2 program = 10 – 30 mg/dL (for monoclonal G, K)
- On HR3 program= 1.5 – 2.0 mg/dL (for albumin and Bence Jones protein)

According to the mobility of the monoclonal component and polyclonal background, the sensitivity may vary.
- Full traceability and positive ID is obtained, by connecting the HYDRASYS to the ASSIST pipetting station.
